Fluorescent proteins as building blocks for genetically encoded biosensors. Principle and -a few- examples

Fluorescent proteins (FP) of the GFP family have become essential tools in imaging over the last 20 years, not only because they allow a specific labeling of proteins compatible with live cell imaging, but also because they gave rise to a large diversity of biosensors to finely visualize, track and quantify cellular events down to the molecular level.

After a short historical introduction, this conference will present in detail the fluorescent proteins and the link between their structure and their optical, chemical and biochemical properties (diversity of color, folding and chromophore maturation, oligomerization tendency, photobleaching, protonation states and isomerization). Then, the different biosensing modalities will be presented, based either on a single FP or on two FPs as a FRET pair. A focus will be done on lifetime detection (FLIM) of the biosensors through a few examples.
