What the transcriptome does not tell – the metabolome is closer to the phenotype

Plants as sessile organisms are highly exposed to injury by mechanical stress and pathogen attack, like herbivore attack. An integrated metabolome and transcriptome analysis of wild type as well as for a jasmonate-deficient mutant plant was performed. To integrate the high amounts of data from both omics technologies the software package MarVis (http://marvis.gobics.de/) was used. Subsets of high quality metabolite and transcript data were finally combined for clustering and visualization by means of one-dimensional self-organizing maps (1D-SOM). This allows pattern comparison of gene expression and metabolite profiles to describe the jasmonate-dependent and jasmonate-independent wound reaction as well as pathways, which are constitutively downregulated in the jasmonate-deficient mutant plant. In particular, our approach also supports the discovery of so far unknown derivatives of the jasmonate biosynthesis. The structure of these compounds was solved by MS/MS experiments.
