The F-box UFO redirects the master floral regulator LEAFY to novel cis-regulatory elements

LEAFY is a floral transcription factor with a key role in most angiosperms. It contributes to conferring a floral fate to meristems. It also helps setting up floral patterning that will result in ABCE genes induction and organ identity determination. LEAFY is expressed uniformly in the floral meristems and how it achieves local induction of some of the ABCE genes is not yet totally understood. LEAFY was show to genetically and physically interact with the protein UFO (Unsual Floral Organs) in various species to perform various actions including the induction of the B genes. UFO is an F-box protein participating in the formation of an SCF E3 Ubiquination complex and thus supposedly targeting some proteins for degradation. Whether LEAFY is truly the target of UFO-mediated ubiquitination and how LEAFY proteolysis would lead to downstream gene activation has remained elusive. By combining plant assays, biochemistry, in vitro genomics and structural biology, we have been able to solve this long-standing question and understand the molecular basis for the LEAFY-UFO synergy.
