Towards the prediction of plant phenotypes under combined abiotic constraints using resource allocation-based models

 

Predicting quantitatively the behavior of living organisms from the finest scales to the individual scale in normal and complex environmental conditions remains highly challenging for the system biology community. Part of the difficulty consists in integrating the scales where the decisions of the adaptation to the environment take place, i.e. the cellular and infra-cellular scales, in the context of the individual. Over the last ten years, a significant step has been achieved in the modeling of cellular and infra-cellular scales in microbial cells. Considering the cell as a set of functional constraints (e.g. biophysical, stoichiometric or osmotic, etc.) falls within the so-called modeling field of constraint-based modeling (CBM) that calls for the resolution of linear optimization problem. We developed and validated experimentally on microbial cells a new CBM method, named Resource Balance Analysis (RBA). RBA predicts for a specific environment, the set of possible cellular configurations (growth rate, metabolic fluxes, abundances of molecular machines, including ribosomes, enzymes, transporters) compatible with the available external resources, and has the potential to predict the cell response to a large set of complex environmental conditions. Recently, the RBA theoretical framework was extended to eukaryotic cells and used to generate a RBA model for the shoot compartment of the plant Arabidopsis thaliana. In this talk, we will present the RBA modeling framework and illustrate its ability to predict plant phenotypes in normal and complex environmental conditions – conditions where environmental abiotic constraints (stresses) combine –
