Expression hétérologue et électrophysiologie végétale
L’IPSiM héberge une plateforme d’expression hétérologue et d’électrophysiologie végétale (EHEV-IPSiM) unique en France. La plateforme regroupe 8 postes d’enregistrements dédiés à des applications spécifiques pour l’expression hétérologue de protéines membranaires de plantes dans des systèmes d’expression cellulaires (ovocytes de xénopes, cellules animales en culture, protoplastes végétaux), et/ou pour des analyses électrophysiologiques (caractérisation d’activité de protéines de transport, essentiellement par voltage-clamp ou patch-clamp sur cellules isolées). Elle est placée sous la responsabilité scientifique d’Anne-Aliénor Véry, DR CNRS et l’Ingénieur en charge de la plateforme est Claire Corratgé-Faillie, IR CNRS.
La plateforme sert aussi de support pour la formation théorique et pratique d’étudiants dans le cadre d’écoles thématiques (« Transport », Univ. Montpellier, école d’électrophysiologie CNRS/INSERM/ Univ. Montpellier « E2M » et l’école internationale bisannuelle « Mistral ») et forme chaque année des jeunes chercheurs à l’électrophysiologie.

Les principaux protocoles pour la préparation des solutions de perfusion, les réglages des étireuses, la programmation des séquences de voltage-clamp sont consignés et accessibles ci-dessous dans l’onglet Protocoles. La gestion des principaux consommables est assurée par la plateforme et la maintenance des matériels est financée sur une ligne spécifique du budget de l’UMR IPSiM. L’agenda de la plateforme (réservation des postes d’enregistrement) est accessible par une application en ligne sur l’Intranet de l’IPSiM (Pour les scientifiques extérieurs, merci de prendre contact avec les responsables).
Un comité des usagers de la plateforme est réuni annuellement par les responsables scientifiques.

Expertises
Expression hétérologue
La plateforme offre des possibilités d’expression hétérologue dans différents contextes :
- Cellules végétales (expression transitoire par « floral dip », « PEG » et « biolistique »)
- Cellules animales (ovocytes de Xénopes, lignées cellulaires animales)
Ces systèmes d’expression sont utilisés à l’IPSiM, principalement pour la caractérisation de protéines membranaires impliquées dans le transport. S’ils offrent la possibilité d’étudier les propriétés fonctionnelles de ces protéines par différents moyens disponibles à l’IPSiM (spectrométrie 15N pour les transporteurs d’azote/acides aminés, utilisation de traceurs radiomarqués ([3H]-ABA/[3H]-IAA) pour les transporteurs d’hormones, gonflement osmotique de protoplastes ou ovocytes pour les aquaporines, …) et sont à ce titre utilisés par pratiquement toutes les équipes de l’IPSiM, ces systèmes d’expression hétérologue sont plus particulièrement exploités dans la seconde composante de la plateforme dédiée aux enregistrements électrophysiologiques.
Electrophysiologie
La plateforme regroupe huit postes d’enregistrement dédiés à des applications spécifiques.
Un premier poste de patch-clamp (Patch-clamp Laser) est équipé d’un laser pour la microdissection de la paroi pecto-cellulosique et permet des enregistrements en patch-clamp in situ sur les poils racinaires (unique en France).
Un deuxième poste de patch-clamp (Patch-clamp Imagerie) est équipé d’un système d’imagerie de la fluorescence (monochomateur + source conventionnelle avec filtres, caméra EMCCD ultra-sensible refroidie) et permet le patch-clamp couplé à l’imagerie par fluorescence (GFP) ou bioluminescence (sonde calcique aequorine).
Ces deux postes sont également disponibles pour des enregistrements en patch-clamp conventionnels, sur protoplaste, cellule animale en culture ou ovocyte de Xénope.
Les six autres postes sont installés dans une grande salle attenante à l’atelier de fabrication des microélectrodes. Deux postes au design identique (Poste Ovocyte 1 et Poste Ovocyte 2) sont dédiés aux enregistrements des forts courants souvent observés dans les ovocytes sur-exprimant des canaux sur ovocyte de Xénope (voltage-clamp à deux électrodes). L’ergonomie de ces deux postes a été optimisée pour en faciliter l’accès à des opérateurs débutants (quelques jours de formation seulement sont nécessaires).
Le troisième poste de patch-clamp (Poste single channel) de la plateforme a été optimisé pour les enregistrements de courants à travers des canaux ioniques isolés (« single channel recording »).
Un autre poste (Poste Signalisation) est dédié aux enregistrements sur système foliaire de plantes entières (Arabidopsis ou plantules de graminées). Il permet l’enregistrement extracellulaire de signaux électriques propagés en réponse à divers stimuli. L’équipement de ce poste permet l’acquisition en différentiel sur 40 canaux différents (électrodes extra-cellulaires), pour enregistrer les signaux électriques (de type potentiel d’action) et cartographier leur propagation dans le système foliaire.
Un poste (Poste Racine) permet les enregistrements intra et extra-cellulaires à l’aide de microélectrodes à potentiel et/ou de microélectrodes ioniques spécifiques (H+, K+, NO3–, …). Le design de ce poste est optimisé pour les racines et permet, par exemple la mesure simultanée de la différence de potentiel et du flux ionique transmembranaires.
Le dernier poste (en construction) sera équipé d’un microscope motorisé spécifique (x320 stéréo et x1000 mono) et permettra les enregistrements intracellulaires de potentiel de membrane ou de concentration ionique.
